报告人简介: Prof. Terence Strick mainly studies the kinetics of multiple DNA repair processes, such as transcription-coupled DNA repair, DNA mismatch repair and non-homologous DNA repair by using single-molecule techniques. He, together with his colleagues, invented the first single-molecule magnetic tweezers in 2002 and this technique has been widely used in the biophysical research field all over the world. In 2004, he developed a single-molecule nanomanipulation technique, which provides single base pair resolution due the property of supercoiled constrained DNA substrates and allows pulse-chase detections with multiple components, and this technique breaks the gate of transcription studies (2006 science) and transcription-coupled DNA repair (2012, 2016 nature). 报告摘要

DNA repair is often a multi-component, multi-step process involving damage recognition, validation of that recogntion step, and damage repair per se. The multistep nature of the process makes it difficult to characterize in detail using standard biochemical techniques. Single-molecule approaches, despite their name, remain powerful tools for reaching deep inside multistep reaction systems to characterize in detail the kinetics and composition of molecular complexes. We will illustrate this in the context of three essential DNA repair systems which are involved in protecting genomes from a range of lesions: DNA mismatch repair, transcription-coupled repair, and non-homologous end-joining.
